FOCUS on Alzheimer’s disease. A recent paper by AA Pieper and colleagues demonstrated that pharmacologic restoration of NAD+ homeostasis with P7C3-A20 reverses cognitive deficits and neuropathology in advanced Aβ- and tau-driven AD models. P7C3-A20 is a novel small molecule NAMPT activator. NAMPT (nicotinamide phosphoribosyltransferase) is the rate-limiting enzyme for NAD+ biosynthesis via the salvage pathway. Neuroprotective and proneurogenic activity has been demonstrated for P7C3-A20 previously in several different models.
