BMS-833923 (1059734-66-5) is a smoothened (SMO) antagonist. It decreased cell proliferation and induced apoptosis (EC50=10 mM) via the intrinsic pathway in esophageal adenocarcinoma cell lines.1 Ameloblastoma cells with an activating SMO-L412F mutation were insensitive to vismodegib (another SMO antagonist) but BMS-833923 significantly reduced downstream Hedgehog signaling and tumor cell viability.2 In developmental studies BMS-833923 has been characterized as a uniquely effective SMO inhibitor when it was used to show that Sonic hedgehog is functionally dedicated to ray branching during fin regeneration in zebrafish.3 Orally active.
