CAIF is a covalent allosteric inhibitor of fucosyltransferase 8 (FUT8; IC50 = 5.7 µM). Selective over FUT3 and FUT4. FUT8 in involved in tumor invasion, metastasis, and immune evasion as well as antitumor immunity and drug resistance. It displayed minimal cytotoxicity and displayed efficacy in inhibiting core fucosylation in multiple cancer cell lines. CAIF also suppressed cancer cell migration and invasion in multiple tumor models.
