CNP7 is a first-in-class covalent inhibitor of HMG-CoA synthase 1, the first enzyme in the mevalonate pathway. Treatment of HCT116 cells with 5 µM CNP7 lead to a significant reduction in HMG-CoA levels and increased levels of unprenylated proteins. It caused cell death in HEK293T and HCT116 cells due to a reduction in geranylgeranyl pyrophosphate levels. An interesting new tool for investigating mevalonate metabolism.
