Ferroptosis suppressor protein-1 (FSP1) along with ubiquinone and NAD(P)H/H+ has been identified as a second ferroptosis suppressing system which prevents lipid peroxidation independently of the cysteine-glutathione-glutathione peroxidase 4 (GPX4) axis.1-3 icFSP1 (1115910-36-5) was discovered in a screen to identify inhibitors of FSP1. However, it does not competitively inhibit FSP1 enzyme activity but triggers subcellular relocalization of FSP1 (at 2.5 µM) from the membrane and FSP1 condensation before ferroptosis induction.  icFSP1-induced FSP1 condensates show droplet-like properties consistent with phase separation. icFSP1-impairs tumor growth and induces FSP1 condensates in tumors in vivo.4
