Olutasidenib (1887014-12-1) is a potent (IC50’s cell-based IDH1 mutants: HCT116-R132H = 21 nM; HCT116-IDH-R132C = 94 nM; U87MG-R132L = 42 nM; U87MG-R132G = 6 nM; U87MG-R132S = 9 nM) and CNS-penetrant mutant isocitrate dehydrogenase 1 inhibitor.1 It induced durable and complete remissions in relapsed or refractory IDH1 mutated acute myeloid leukemia.2,3
