Prexasertib (1234015-54-3) is a potent inhibitor of CHK1 (IC50 = <1 nM) and CHK2 (IC50 = 8 nM).1 It also inhibits RSK1 (IC50 = 9 nM).1 Prexasertib displayed efficacy in various cancer models alone and in combination with other agents including anti-PD-L1 therapy.2-6 It promoted neuroprotection, axon regeneration, and functional recovery after CNS injury.7 The cytotoxic effects of prexasertib have been attributed to dual inhibition of CHK1 and AMPK.8
