In a paper just out in Redox Biology, the authors show that inhibition of 15-OH-PGDH with SW033291 restores redox homeostasis and confers neuroprotection in Parkinson’s disease. 15-OH-PGDH oxidizes PGE2 to 15-keto PGE2 rendering it biologically inactive. Inhibition of this metabolic pathway with SW033291 (or using genetic methods) mitigates microglial and astrocyte activation, dopaminergic neuron loss, and motor impairment in three mouse models of PD.
