RPI-GLYT2-82 is a reversible, allosteric antagonist (IC50 = 554 nM) of Glycine Transporter 2(GLYT2) with selectivity over GLYT1. It is also an antagonist of the serotonin receptor 5-HT2A (IC50 = 1.9 µM). RPI-GLYT2-82 displayed efficacy in the CCI and PSNL mouse models of neuropathic pain. It showed no addiction liability and had no on-target side effects.
