Y-320 (288250-47-5) is an inhibitor of IL-17 production (IC50 = 20 – 60 nM in CD4 T cells).1 It significantly inhibited the development and progression of arthritis and joint destruction in collagen-induced arthritis models. Y-320 acted synergistically with other chemotherapeutic agents to overcome multidrug resistance.2  It also enhances premature termination codon (PTC) readthrough resulting in increased protein levels and protein synthesis.3 Y-320 also acts as a potent oral sialylation inhibitor (IC50 = 200 nM in PDAC cells).4 It suppressed α-2,3/2,6-sialylation inhibiting PDAC tumor growth and remodeling the tumor microenvironment.  Y-320 acted synergistically with anti-PD-1 therapy to overcome immune checkpoint blockade resistance.
