FOCUS on Drug Repurposing. Montelukast is a cysteinyl leukotriene receptor (CysLTR1) antagonist currently in use for asthma. In a paper just out in Nature Cancer, a group at the Lurie Comprehensive Cancer Center demonstrated that CysLTR1 sustains tumor-promoting myelopoiesis. Genetic ablation of CysLTR1, as well as pharmacological antagonism with montelukast, diminished tumor growth and enhanced antitumor immunity. Targeting CysLTR1 with montelukast overcomes resistance to anti-PD1 in multiple models. These results suggest a new pharmacological strategy to improve cancer immunotherapy.
