FOCUS on Stem Cell Rejuvenation. In the current issue of Cell Stem Cell, a group from Stanford reported that an endogenous eicosanoid, prostaglandin E2 (PGE2) reversed aged muscle stem cell dysfunction leading to increased regeneration and strength in mice.
In a related development, Zhang et al. and Shu et al. showed that blocking metabolism of PGE2 with SW033291 ameliorated age-related muscle atrophy and heart failure.
PGE2 is inactivated by oxidation to the 15-Keto metabolite via 15-Hydroxyprostaglandin dehydrogenase (15-PGDH) which is potently inhibited by SW033291, thus prolonging the half-life of endogenously released PGE2 in vivo.
